Delayed Contralateral Nephrectomy Halted Post-Ischemic Renal Fibrosis Progression and Inhibited the Ischemia-Induced Fibromir Upregulation in Mice

(1) Background: Ischemia reperfusion (IR) is the leading cause of acute kidney injury (AKI) and results in predisposition to chronic disease. We demonstrated that delayed contralateral nephrectomy (Nx) greatly improved function IR-injured decelerated fibrosis progression. Our aim was identify microRNAs (miRNA/miR) involved this process. (2) Methods: NMRI mice were subjected 30 min renal IR one week later Nx/sham surgery. The experiments conducted for 7–28 days after IR. On day 8, multiplex miRNA profiling performed. Expression nine miRNAs determined with qPCR at all time points. Based on target prediction, plexin-A2 Cd2AP measured by Western blot. (3) Results: 8 IR, expression 20/1195 doubled, 9/13 selected upregulated Nx reduced several ischemia-induced pro-fibrotic (fibromirs), such as miR-142a-duplex, miR-146a-5p, miR-199a-duplex, miR-214-3p miR-223-3p, injured kidneys various Plexin-A2 10, while unchanged. (4) Conclusion: progression decreased fibromirs. protein mainly regulated factors other than miRNAs.
